Abstract:

Identification of the driver mutations in cancer has resulted in the development of a new categoryWhile most cancer therapies are given in combinations that are designed to avoid drug resistance, weThis approach is based on notion that drug resistance comes at a fitness cost to the cancer cell thatWe discuss the development of sequential drug therapies in which the first therapy is not given withcurative intent, but to induce a major new sensitivity that can be targeted with a second drug that selectively

Bioorthogonal chemistry based on-demand drug delivery system in cancer therapy

Frontiers of Chemical Science and Engineering 2023, Volume 17, Issue 4,   Pages 483-489 doi: 10.1007/s11705-022-2227-2

Abstract: Benefiting from the advantage of taking place in biological environments without interfering with an innate biochemical process, the bioorthogonal reaction that commonly contains the “bond formation” and “bond cleavage” system has been widely used in targeted therapy for a variety of tumors. Herein, several prominent cases based on the bioorthogonal reaction that tailoring the metabolic glycoengineering tactics to modified cells for cancer immunotherapy, and the innovative tactics for reducing the metal ions’ toxic and side effects with microneedle patches will be highlighted. Based on these applications, the complexities, potential pitfalls, and opportunities of bioorthogonal chemistry in future cancer therapy will be evaluated.
